Master’s Thesis at the University of Kerbala on the Use of Tissue Culture Technology in the Propagation of Local Lemon (Citrus limon)

A Master’s thesis was defended at the College of Agriculture, University of Kerbala, on employing tissue culture technology along with certain physical and chemical stimulants in the propagation of the local lemon (Citrus limon).

The objectives of the thesis were to:
• Select the best combination of growth regulators to achieve the initiation stage of local lemon plants.
• Determine the optimal concentrations of melatonin and the best light source for establishing shoot cultures.
• Study different combinations of growth regulators for rooting of shoots and test the best medium strength for root induction in shoots.

The thesis was submitted by the student Majid Abbas Habib Hussein.

The study concluded that relying on this technique helps preserve local cultivars from extinction or genetic degradation, and that tissue culture can serve as an effective method for propagating rare or local citrus varieties.
